OP, if you are switching from run flats to standard tires, you should purchase a plug kit and keep it in the trunk. That or a can of Fix-a-flat is about all you can do outside of buying a wheel from a salvage yard, mounting a tire on and keeping it in your trunk.

And no, a screw will not do the trick. Maybe with a tiny puncture, in a pinch, with no other alternative. ....Get a plug kit to keep you on the road and then take the tire to get a proper patch applied to the inside.
